氣(qi)相色(se)譜(pu)六(liu)通(tong)閥(fa)是由圓(yuan)形密(mi)封墊(轉子(zi))和固(gu)定(ding)底(di)座(定子(zi))組成。當在充樣(Load)位置時(shi),從進樣(yang)孔(kong)充樣進定(ding)量(liang)環,多余(yu)樣(yang)品從放(fang)空孔(kong)排(pai)出(chu);轉動至進樣(yang)(Inject)位置時(shi),由泵輸送的(de)流(liu)動相(xiang)沖(chong)洗(xi)定(ding)量(liang)環,推動樣(yang)品(pin)入(ru)柱。六(liu)通(tong)閥(fa)較為通(tong)用(yong),各(ge)大(da)HPLC儀器(qi)制造(zao)商(shang)均(jun)以此(ci)產(chan)品作(zuo)為(wei)儀(yi)器(qi)的進樣(yang)器(qi)。
氣(qi)相色(se)譜(pu)六(liu)通(tong)閥(fa)的取(qu)樣方(fang)式(shi)和取(qu)樣工具(ju):
取(qu)樣方(fang)式(shi):為防止可(ke)能(neng)造(zao)成的環境中(zhong)的氣(qi)體成分對(dui)樣(yang)品的汙染或(huo)幹擾(rao),通(tong)過(guo)大(da)註射(she)器(qi)針頭(tou)象(xiang)液體進樣(yang)壹(yi)樣打(da)入(ru)定(ding)量(liang)管。不易(yi)用(yong)各(ge)種膠(jiao)管或塑(su)料(liao)管接(jie)入(ru)這(zhe)可(ke)能(neng):
a)管材本身(shen)不純(chun)凈;
b)各種管材原則上講都(dou)會(hui)有滲透(tou)作用(yong),這(zhe)對(dui)痕(hen)量(liang)分析(xi)尤(you)其(qi)不利。
取(qu)樣工具(ju):目(mu)前常(chang)用(yong)的(de)是金屬鍍膜取(qu)氣(qi)袋、大(da)註射(she)器(qi)或專(zhuan)用(yong)取(qu)氣(qi)鋼(gang)瓶。除(chu)非要(yao)求(qiu)極低,目前已(yi)很(hen)少(shao)采用(yong)球(qiu)膽(dan)、塑(su)料(liao)袋取(qu)氣(qi)等;
定(ding)量(liang)管內樣品的氣(qi)壓:由於(yu)氣(qi)體的(de)含(han)量(liang)和氣(qi)壓直接(jie)有關,為(wei)保證每次進樣(yang)的(de)重復(fu)性(xing),取(qu)樣後(hou)要使定量(liang)管的壓力與大(da)氣(qi)壓平衡,依據經驗壹(yi)般在取(qu)樣後(hou)平衡20~30秒即可(ke);
沖(chong)洗(xi)定(ding)量(liang)管樣品體積(ji):由於(yu)被(bei)分(fen)析(xi)的(de)氣(qi)體樣(yang)品濃(nong)度(du)不同(tong),為(wei)防止進較(jiao)高濃(nong)度(du)後(hou)又進較(jiao)低濃(nong)度(du)樣(yang)品時(shi),定量(liang)管中原有高濃(nong)度(du)氣(qi)體殘(can)留(liu)的幹擾(rao)。取(qu)樣時(shi)要求(qiu)用(yong)新(xin)樣品(pin)氣(qi)對(dui)定(ding)量(liang)管進行(xing)沖(chong)洗(xi),沖(chong)洗(xi)氣(qi)量(liang)依據經驗不小(xiao)於(yu)定(ding)量(liang)體積(ji)的5倍。實際影響也可(ke)以通(tong)過(guo)實(shi)驗(yan)峰的重(zhong)現(xian)性(xing)來判(pan)斷與選擇;
